S&M room and the "secret" room is the same room btw.
Technically the best way for a room to be soundproofed is to be use several noise cancelling materials and to vacuum seal the room to make it air-tight. However as far as I know, noise canceling materials aren't 100%. Plus, vacuum sealing requires a motor - which is definitely not silent - as well as taking time - something Asami and Akihito didn't have. So at best, Asami and Akihito are in a room that cancels most sound.
The other option is that there is a proper panic room that we haven't seen somewhere.
Why should they make noise when a bunch of men are on the other side of the room looking to kill them?
i think the secret room is the room that Akihito accidently found it when he cleaning the masterbedroom from the previous chapter where he found the antique gun and throw it away...or maybe the S&M room where Asami punish Akihito with bondage..
