Glasses guy and the bottom are fated mates. The bottom rejected ml's pheromones after which doesnt rlly make sense since he shouldve rejected them since the beginning but yeah
It's possible he's rejecting it now after meeting his fated which triggered something in his body cause his body kept wanting Kira but his mind didn't. Plus Sakuma was in heat so his senses are heightened from soothing pheromones of his fated to another Alpha's.
The heat part doesn't make sense because I'm pretty sure they have interacted while he was in heat (I haven't read this in so long) but I was thinking it could be because of your first point
