He was just watching his son fuck the guy he claims to love. That's why he came back and wants Set to come to Duat with him because this mother fucker feels like he can only do that to Set. I think Osiris doesn't know who fucked Seth. Remember that Horus is not a god yet so the other gods of Ennead cannot detect their specific presence (only gods can detect each others presence). Osiris got an idea that Seth had sex because of the hickeys and he pierced Seth's abdomen to confirm if he got inseminated and basically confirmed it when the vines did get semen. He doesn't know who specifically had sex with Seth, because if he did know it's Horus, I don't think he'd sit around and turn a blind eye. He'll probably kill Horus, imo.
