I think it's more on wanting to create a new species of demon like Inna lab where he can modify it and stuff, ameri was made biologically and naturally but I'm guessing the dad wants to create his own demon
The substitute demon lord might've made a deal with him so that he wouldn't touch Henry and Ameri

wait im curious, if he needs human blood and ameri js half human... well im guessing her blood doesn't make demons react like the way with pure human blood BUT isn't ameri's existence already an answer to his research?
